Mg2+ mediates interaction between the voltage sensor and cytosolic domain to activate BK channels.

چکیده
The voltage-sensor domain (VSD) of voltage-dependent ion channels and enzymes is critical for cellular responses to membrane potential. The VSD can also be regulated by interaction with intracellular proteins and ligands, but how this occurs is poorly understood. Here, we show that the VSD of the BK-type K(+) channel is regulated by a state-dependent interaction with its own tethered cytosolic domain that depends on both intracellular Mg(2+) and the open state of the channel pore. Mg(2+) bound to the cytosolic RCK1 domain enhances VSD activation by electrostatic interaction with Arg-213 in transmembrane segment S4. Our results demonstrate that a cytosolic domain can come close enough to the VSD to regulate its activity electrostatically, thereby elucidating a mechanism of Mg(2+)-dependent activation in BK channels and suggesting a general pathway by which intracellular factors can modulate the function of voltage-dependent proteins.

Question: Do Mg2+ and Ca2+ work through functionally similar mechanisms to activate BK channels? Background: BK channels are large conductance Ca2+ and voltage-activated K+ channels, which allow K+ to leave the cytoplasm and promote membrane hyperpolarization under physiological conditions when activated by membrane potential and/or intracellular Ca2+. In addition to these two primary signals, ...
متن کاملInteraction between residues in the Mg2+-binding site regulates BK channel activation
As a unique member of the voltage-gated potassium channel family, a large conductance, voltage- and Ca(2+)-activated K(+) (BK) channel has a large cytosolic domain that serves as the Ca(2+) sensor, in addition to a membrane-spanning domain that contains the voltage-sensing (VSD) and pore-gate domains. The conformational changes of the cytosolic domain induced by Ca(2+) binding and the conformat...
متن کاملLarge conductance Ca2+-activated K+ (BK) channel: activation by Ca2+ and voltage.
Large conductance Ca2+-activated K+ (BK) channels belong to the S4 superfamily of K+ channels that include voltage-dependent K+ (Kv) channels characterized by having six (S1-S6) transmembrane domains and a positively charged S4 domain. As Kv channels, BK channels contain a S4 domain, but they have an extra (S0) transmembrane domain that leads to an external NH2-terminus. The BK channel is activ...
متن کاملMg2+-dependent Regulation of BK Channels: Importance of Electrostatics
A signature feature of large conductance, calcium-and voltage-activated K + channels (now usually termed " BK " channels) is their dual regulation by two physiological signals, cytosolic Ca 2+ and membrane voltage. This dual-sensing capacity of BK channels distinguishes them from other voltage-dependent K + channels in terms of the physiological roles they can play, allowing their voltage-sensi...
متن کاملMolecular mechanism of pharmacological activation of BK channels.
Large-conductance voltage- and Ca(2+)-activated K(+) (Slo1 BK) channels serve numerous cellular functions, and their dysregulation is implicated in various diseases. Drugs activating BK channels therefore bear substantial therapeutic potential, but their deployment has been hindered in part because the mode of action remains obscure. Here we provide mechanistic insight into how the dehydroabiet...
